Identifying local and descending inputs for primary sensory neurons.

چکیده
Primary pain and touch sensory neurons not only detect internal and external sensory stimuli, but also receive inputs from other neurons. However, the neuronal derived inputs for primary neurons have not been systematically identified. Using a monosynaptic rabies viruses-based transneuronal tracing method combined with sensory-specific Cre-drivers, we found that sensory neurons receive intraganglion, intraspinal, and supraspinal inputs, the latter of which are mainly derived from the rostroventral medulla (RVM). The viral-traced central neurons were largely inhibitory but also consisted of some glutamatergic neurons in the spinal cord and serotonergic neurons in the RVM. The majority of RVM-derived descending inputs were dual GABAergic and enkephalinergic (opioidergic). These inputs projected through the dorsolateral funiculus and primarily innervated layers I, II, and V of the dorsal horn, where pain-sensory afferents terminate. Silencing or activation of the dual GABA/enkephalinergic RVM neurons in adult animals substantially increased or decreased behavioral sensitivity, respectively, to heat and mechanical stimuli. These results are consistent with the fact that both GABA and enkephalin can exert presynaptic inhibition of the sensory afferents. Taken together, this work provides a systematic view of and a set of tools for examining peri- and extrasynaptic regulations of pain-afferent transmission.

Rapid arrival and integration of ascending sensory information in layer 1 nonpyramidal neurons and tuft dendrites of layer 5 pyramidal neurons of the neocortex.
Ascending sensory inputs arriving in layer 1 of the neocortex carry crucial signals for detecting salient information; but how the inputs are processed in layer 1 is unknown. Using a whole-cell in vivo recording technique targeting nonpyramidal neurons in layer 1 and tuft dendrites of layer 5 pyramidal neurons in layers 1-2, we examined the processing of these ascending sensory inputs in the ba...
متن کاملConvergence of Cortical and Sensory Driver Inputs on Single Thalamocortical Cells
Ascending and descending information is relayed through the thalamus via strong, "driver" pathways. According to our current knowledge, different driver pathways are organized in parallel streams and do not interact at the thalamic level. Using an electron microscopic approach combined with optogenetics and in vivo physiology, we examined whether driver inputs arising from different sources can...
متن کاملResponses of Drosophila giant descending neurons to visual and mechanical stimuli.
In Drosophila, the paired giant descending neurons (GDNs), also known as giant fibers, and the paired giant antennal mechanosensory descending neurons (GAMDNs), are supplied by visual and mechanosensory inputs. Both neurons have the largest cell bodies in the brain and both supply slender axons to the neck connective. The GDN axon thereafter widens to become the largest axon in the thoracic gan...
متن کاملA two-stage unsupervised learning algorithm reproduces multisensory enhancement in a neural network model of the corticotectal system.
Multisensory enhancement (MSE) is the augmentation of the response to sensory stimulation of one modality by stimulation of a different modality. It has been described for multisensory neurons in the deep superior colliculus (DSC) of mammals, which function to detect, and direct orienting movements toward, the sources of stimulation (targets). MSE would seem to improve the ability of DSC neuron...
متن کاملMorphological Identification of Cell Death in Dorsal Root Ganglion Neurons Following Peripheral Nerve injury and repair in adult rat
Background: Axotomy causes sensory neuronal loss. Reconnection of proximal and distal nerve ends by surgical repair improves neuronal survival. It is important to know the morphology of primary sensory neurons after the surgical repair of their peripheral processes. Methods: Animals (male Wistar rats) were exposed to models of sciatic nerve transection, direct epineurial suture repair of sciati...
